玉米自交系×大刍草远缘杂交后代性状变异研究初探   总被引:5,自引:2,他引:3  
采用人工剥苞多次授粉方法,将近缘一年生二倍体类玉米(大刍草)与普通玉米自交系杂交,对F1、F2以及回交后代B1的抽雄期、吐丝期、散粉期、株高、穗位高、分蘖进行调查研究,分析远缘杂交后代农艺性孳的遗传变化。结果表明,远缘杂交后代株高、穗化高分离变异较大,而抽雄期、吐丝期、散粉期分离变异较小,不同世代植株分蘖变化较大,F1代有57.5%植株、F2代有23%植株、BC1F1代有0.8%植株有分蘖;雌穗光周期敏感性大于雄穗。  相似文献

以一粒小麦-葡萄牙野燕麦杂交后代一粒葡为实验材料,以地高辛标记的葡萄牙野燕麦基因组DNA为探针、一粒小麦基因组DNA为封阻对一粒葡及其衍生系根尖染色体进行基因组原位杂交(GISH)分析,探讨了影响一粒葡GISH效果的主要因素.建立并优化了一粒葡GISH分析的实验体系,即探针DNA与封阻DNA比例为1∶50时可有效分开双方染色体组.优化GISH分析显示,在一粒葡后代衍生系中均检测到燕麦染色质的存在,且不同选系间带有燕麦染色体的数目不同,进一步证明一粒葡是一粒小麦-葡萄牙野燕麦远缘杂交的后代.  相似文献

Delineation of the relationship between sequence and structure in proteins has proven elusive. Most studies of this problem use alignment methods and other approaches based on the characteristics of individual residues. It is demonstrated herein that the sequence‐structure relationship is determined in significant part by global characteristics of sequence organization. Information encoded in complete sequences is required to distinguish proteins in different architectural groups. It is found that the statistically significant differences between sequences encoding different architectures are encoded in a surprisingly small set of low‐wave‐number sequence periodicities. It would therefore appear that unexpected simplicity in an appropriately defined Fourier space may be an inherent characteristic of the sequences of folded proteins. Genomic imprinting is a common epigenetic phenomenon in mammals. Dysregulation of genomic imprinting has been implicated in a variety of human diseases. ZFP57 is a master regulator in genomic imprinting. Loss of ZFP57 causes loss of DNA methylation imprint at multiple imprinted regions in mouse embryos, as well as in embryonic stem (ES) cells. Similarly, mutations in human ZFP57 result in hypomethylation at many imprinted regions and are associated with transient neonatal diabetes and other human diseases. Mouse and human Zfp57 genes are located in the same syntenic block. However, mouse and human ZFP57 proteins only display about 50% sequence identity with different number of zinc fingers. It is not clear if they share similar mechanisms in maintaining genomic imprinting. Here we report that mouse and human ZFP57 proteins are functionally interchangeable. Expression of exogenous wild-type human ZFP57 could maintain DNA methylation imprint at three imprinted regions in mouse ES cells in the absence of endogenous mouse ZFP57. However, mutant human ZFP57 proteins containing the mutations found in human patients could not substitute for endogenous mouse ZFP57 in maintaining genomic imprinting in ES cells. Like mouse ZFP57, human ZFP57 and its mutant proteins could bind to mouse KAP1, the universal cofactor for KRAB zinc finger proteins, in mouse ES cells. Thus, we conclude that mouse and human ZFP57 are orthologs despite relatively low sequence identity and mouse ES cell system that we had established before is a valuable system for functional analyses of wild-type and mutant human ZFP57 proteins.  相似文献

A short review of main cytogenetic features of insects belonging to the sister neuropteran families Myrmeleontidae (antlions) and Ascalaphidae (owlflies) is presented, with a particular focus on their chromosome numbers and sex chromosome systems. Diploid male chromosome numbers are listed for 37 species, 21 genera from 9 subfamilies of the antlions as well as for seven species and five genera of the owlfly subfamily Ascalaphinae. The list includes data on five species whose karyotypes were studied in the present work. It is shown here that antlions and owlflies share a simple sex chromosome system XY/XX; a similar range of chromosome numbers, 2n = 14-26 and 2n = 18-22 respectively; and a peculiar distant pairing of sex chromosomes in male meiosis. Usually the karyotype is particularly stable within a genus but there are some exceptions in both families (in the genera Palpares and Libelloides respectively). The Myrmeleontidae and Ascalaphidae differ in their modal chromosome numbers. Most antlions exhibit 2n = 14 and 16, and Palparinae are the only subfamily characterized by higher numbers, 2n = 22, 24, and 26. The higher numbers, 2n = 20 and 22, are also found in owlflies. Since the Palparinae represent a basal phylogenetic lineage of the Myrmeleontidae, it is hypothesized that higher chromosome numbers are ancestral for antlions and were inherited from the common ancestor of Myrmeleontidae + Ascalaphidae. They were preserved in the Palparinae (Myrmeleontidae), but changed via chromosomal fusions toward lower numbers in other subfamilies.  相似文献

系统发育关系的构建对被子植物分类及进化研究非常重要。长期以来,被子植物系统发育的研究,大多使用质体基因、线粒体基因或少数保守的单拷贝核基因。该研究从已注释基因组或转录组中搜集88种被子植物(包含58目)的核基因集;通过对其进行同源基因聚类及去旁系同源基因,获得了5 993个一对一的直系同源基因家族(即对于每个基因家族,每种植物最多一条序列,最少包含50个物种);使用截取各种不同数目基因集的DNA或氨基酸序列,采用串联法(concatenation)和溯祖法(coalescence),共构建了20棵进化树。比较这些进化树,虽然大部分结果支持APG IV中描述的被子植物主要支系之间的关系[(真双子叶植物,单子叶植物),木兰类植物],但真双子叶植物内部各目分支的演化关系与APG IV有一个很大的不同,即认为檀香目和石竹目是蔷薇类植物的姊妹群。基于这些进化树,估算了被子植物各目分支的分化时间,结果表明被子植物的起源时间为237.78百万年前(95%置信区间为202.6~278.08),与主流观点认为的225百万年至240百万年前一致。以上结果为构建进化树提供了一种可行性策略,这种方法允许使用基因数目更多而计算速度更快。  相似文献

The ability to move from the primary infection site and colonize distant tissue in the leaf is an important property of bacterial plant pathogens, yet this aspect has hardly been investigated for model pathogens. Here we show that GFP‐expressing Pseudomonas syringae pv. syringae DC3000 that lacks the HopQ1‐1 effector (PtoDC3000ΔhQ) has a strong capacity to colonize distant leaf tissue from wound‐inoculated sites in N. benthamiana. Distant colonization occurs within 1 week after toothpick inoculation and is characterized by distant colonies in the apoplast along the vasculature. Distant colonization is blocked by the non‐host resistance response triggered by HopQ1‐1 in an SGT1‐dependent manner and is associated with an explosive growth of the bacterial population, and displays robust growth differences between compatible and incompatible interactions. Scanning electron microscopy revealed that PtoDC3000ΔhQ bacteria are present in xylem vessels, indicating that they use the xylem to move through the leaf blade. Distant colonization does not require flagellin‐mediated motility, and is common for P. syringae pathovars that represent different phylogroups.  相似文献

中国玉米种质资源少,只有远缘杂交才有可能获得遗传多样性。但是,远缘杂交F2的疯狂分离,使得一个优良性状的稳定往往需要经过多代选择,耗时费力。我们利用从孤雌生殖产生的纯合2倍体,仅2~3年即选育出了优良异源种质纯系540(Fig.1),而常规育种至少要5~6年。本工作选用zi330,旋9,MO17,黄早4等自交系与远缘种属2倍体多年生类玉米(DP)杂交,将其F1的BC1去雄,在花丝上喷撒MH,DMSO,PEG,诱导孤性生殖。纯系540的株高、穗位、穗长等农艺性状(Fig.3)优于DP(Fig.2),但其抗逆性优于亲本zi330。染色体分带结果:纯系540(Fig.6)的总带数为8,与亲本(Fig.4&5)相同。但其C带特征,一方面,兼有二者的特点,如:第2对同源染体的一条染色体长臂,和第5对同源染色体的两条长臂均表现DP的C带特征,为异染色质末端带;另一方面,出现两者之间没有的现象,如:第8对同源染色体的长臂近末端显示异染色质带,为同型配对,而zi330的笫8对同源体不显带。这可能是染色体重组的结果。  相似文献
